Intro:
There is two main purposes of this lab. The first being to gain lab experience and to be better at lab procedures. The second purpose was to use our DNA to learn and work with PCR and gel electrophoresis. My hypothesis was that through the procedure I would see my alu insert and see the different genotypes I have. This would only be able to happen if the lab was done exactly correct. If it would have worked we could have written up and correct report to state our results.
Data:
My results are on line 1, column c.
Genotype Chart
Possible Bands Allele Designation Genotype Alu Insert
One band at 415 bp -/- homozygous No Alu insert
One band at 715 bp +/+ homozygous Alu insert present on both chromosomes
One band at 415 bp and a second band at 715 bp +/- heterozygous Alu insert on one of the chromosomes
This table explains the results that you can get different results. Based on these results you can figure out your alu insert and genotype. This lab had to be very precise to get the right outcome. Unfortunately because we did not get any results I don't have a result to see what my alu insert or genotype is. During the lab I had to re centrifuged my dna because I sucked it up into my micropipette on accident. That might have thrown off the results that I got.
Observations
Day 1:
~ Today I saw that there is a very specific measurement of materials that has to go in with the DNA.
Day 2:
~Today I saw that when we laid the materials in the ice box it was very difficult to put the material in and be able to not spill the vial.

Biology Concepts
PCR: Stands for Polymerase Chain Reaction. It is used to duplicate a small section of DNA
Gel Electrophorisis: A process that separates DNA by long and short strands
Allele: A form of a gene from mutation.

Conclusion
Overall this lab was inconclusive. Do to the fact that the lab had no evidence we could not come to any conclusion during this project. I gained a lot of experience from this lab.During this lab we worked on using pipettes and using the different lab equipment for future labs. Since we didn't get any results this lab, we can’t come to any conclusion. If we had come to a conclusion we would have figured out the alu insert and genotype.
